FUNGAL SEROLOGY

TEST REAGENTS FOR SERODIAGNOSIS:

These immunodiffusion reagents are used to detect antibodies in patient's sera with histoplasmosis, blastomycosis, andcoccidioidomycosis. Antigen and antisera may be purchased together in a kit with immunodifflision plates or separately. Kits contain 1.0 ml of antigen and 2.0 ml of antisera, a sufficient quantity for 50 patterns (maximum 200 tests), & 5 immunodiffusion plates, each containing 7 patterns. The reagents may be used with either the macro or micro patterns. Tubed ID Agar may be substituted for plates upon request (two tubes for each plate).

TEST REAGENTS FOR SERODIAGNOSIS OF CANDIDIOSIS

These immunodiffusion (ID) reagents are used to detect antibodies in a patient with candididosis*. A positive ID test may indicate invasive colonization of a patient by a Candida spp. or T. Glabrata. The reaction between control antigen and antiserum should give at least two to three precipitation bands. When reporting an ID test, bands of identity constitute a positive test, however, bands of partial or non-identity are suspicious and indicate additional studies are necessary. Kits contain 1.0 ml of antigen and 2.0 ml of antiserum, a sufficient quantity for 50 patterns (200 tests) and 5 immunodiffusion plates each containing 7 patterns.

TEST REAGENTS FOR SERODIAGNOSIS OF ASPERGILLOSIS

These immunodiffusion (ID) reagents are used to detect antibodies in patients with allergic bronchopulmonary aspergillosis, aspergilloma (fungus ball), and invasive aspergillosis. Antigens and antisera may be purchased in a kit with 10 tubes of agar to prepare 10 microimmunodiffusion plates or separately (microimmunodiffusion templates are available from Quality House). Microimmunodiffusion plates are recommended for the Aspergillosis ID test since the bands are sharper and have better separation than macro ID patterns.
Kits contain 1.0 ml each of antigen (A. flavus, A. fumigatus, & A. niger) and 2.0 ml of antisera (A. flavus, A. fumigatus & A. niger), a sufficient quantity for 150 patterns (50 patterns with A. flavus, 50 patterns with A. fumigatus and 50 patterns with A. niger), a maximum of 200 tests can be run with each 3 sets of reagents, and 10 tubes of ID agar (enough to pour 10 ID plates). When performing the ID test for aspergillosis, line(s) of identity by a patient's serum with references indicates a positive reaction. Line(s) of partial or nonidentity are considered suspicious and indicate further studies are necessary. Some line(s) of non-identity may be due to C-reactive protein in a patient's serum and may be eliminated by soaking the agar with sodium citrate for one hour. It is recommended that patients suspected of having aspergillosis be run against all three aspergillus control systems.

TEST REAGENTS FOR SEROLOGICAL IDENTIFICATION (EXOANTIGEN)

These immunodiffuision agents are used for the serological identification of cultures of blastomyces dermatitidis, Coccidioides immitis, and Histoplasma capsulatum. Antigens and antisera may be purchased together in a kit with immunodiffusion plates or separately. Large kits contain 2.0 ml of antigen and 1.0 ml of antiserum, a sufficient quantity for 50 patterns (maximum of 100 tests) and 5 immunodiffuision plates, each containing 7 patterns. A maximum of 2 unknown culture extracts can be tested in each pattern, one in duplicate wells on either side of the control antigens. The reagents may be used with either the macro or micro patterns. Tubed ID Agar may be substituted for plates upon request (two tubes for each plate).
