USE
Oxidase
Swab is a convenient, stable oxidase reagent swab for use
in the presumptive identification of Neisseria and
as an aid in the identification of Pseuaomonas and
other cytochrome oxidase-positive gram negative microorganism
SUMMARY
AND PRINCIPLES
In some
bacteria, electrons are transferred to cytochrome oxidase
before being transferred to oxygen in aerobic respiration.
The presence of cytochrome oxidase is used to identify these
bacteria.
FORMULA
Active
Ingredient: phenylenediamine
This
formulation is typical. Production lots may be adjusted,
if necessary1,to offset variations Is raw
materials in order to meet performance criteria.
PRECAUTIONS
This
mediurn is for IN VITRO DIAGNOSTIC USE only Precautions
should be taken against the dangers of microbiological hazards.
Specimens, containers, and media should be sterilized after
use.
STORAGE
INSTRUCTIONS
Oxidase
swabs should be stored at room temperature in the dark and
remain in the original packaging until needed.
EVIDENCE
OF DETERIORATION
This
product should not be used if blue discoloration appears
on swabs. If any deficiencies of these types are observed,
notify the manufacturer.
PROCEDURE
Touch
swab to colony on culture medium. Examine for purple to
black coloration after 10 seconds
(oxidase
positive). Disregard any color development occurring after
60 seconds

Test
lot samples with stock cultures of Neisseda gonorrhoea.
and Eschedchia coil.
N.
gonorrhoeae 43069 : Purple to Black color change
E. coli
25922 : No Purple color
LIMITATION
OF THE PROCEDURES
Do not
perform oxidase test on cultures from medium containing
Defibrinated Sheep Blood, may give false positives. Further
testing is required to identify the colonies. Biochemical
and serological procedures may be found in appropriate references.
